The phosphoenolpyruvate-initiated pathway of fructose metabolism in Escherichia coli.

چکیده

The inability of a particular Escherichiu coZi mutant to form colonies on fructose minimal medium is ascribed to the inhibition of growth by glucose 6-phosphate, which accumulates in this strain from a small amount of glucose in the fructose. This explanation requires that fructose metabolism not be initiated by its phosphorylation to fructose 6-phosphate. Experiments in vitro show that fructose is phosphorylated to fructose l-phosphate in a reaction catalyzed by the phosphoenolpyruvate phosphotransferase system. Fructose l-phosphate is converted to fructose 1,6-diphosphate by an inducible kinase, as has been found in other organisms. Thus, fructose should be considered a gluconeogenic carbon source in E. coli.

Fructose and mannose metabolism in Aeromonas hydrophila: identification of transport systems and catabolic pathways.

Aeromonas hydrophila was examined for fructose and mannose transport systems. A. hydrophila was shown to possess a phosphoenolpyruvate (PEP): fructose phosphotransferase system (fructose-PTS) and a mannose-specific PTS, both induced by fructose and mannose. The mannose-PTS of A. hydrophila exhibited cross-reactivity with Escherichia coli mannose-PTS proteins. The fructose-PTS proteins exhibited...

Enhancing succinic acid biosynthesis in Escherichia coli by engineering its global transcription factor, catabolite repressor/activator (Cra)

This study was initiated to improve E. coli succinate production by engineering the E. coli global transcription factor, Cra (catabolite repressor/activator). Random mutagenesis libraries were generated through error-prone PCR of cra. After re-screening and mutation site integration, the best mutant strain was Tang1541, which provided a final succinate concentration of 79.8 ± 3.1 g/L: i.e., 22....

Novel allosteric activation site in Escherichia coli fructose-1,6-bisphosphatase.

Fructose-1,6-bisphosphatase (FBPase) governs a key step in gluconeogenesis, the conversion of fructose 1,6-bisphosphate into fructose 6-phosphate. In mammals, the enzyme is subject to metabolic regulation, but regulatory mechanisms of bacterial FBPases are not well understood. Presented here is the crystal structure (resolution, 1.45A) of recombinant FBPase from Escherichia coli, the first stru...

Inducible phosphoenolpyruvate-dependent hexose phosphotransferase activities in Escherichia coli.

1. A method is described for measuring the rate of phosphoenolpyruvate-dependent phosphotransferase activity for a variety of hexoses in toluene-treated suspensions of Escherichia coli. 2. The specific activities of the phosphotransferases that catalyse the phosphorylation of hexoses are greatly affected by the carbon source for growth. 3. In all strains of E. coli tested, fructose phosphotrans...
